The Effect of Tinzaparin Treatement on BAX and CCNB1 Gene Expression in an Oral Squamous Cell Carcinoma Nude Mice Model

Introduction: This study aims to evaluate the effect of tinzaparin treatment on the expression of proapoptotic BAX and cell cycle G2/M phase regulator CCNB1 genes in an oral squamous cell carcinoma (OSCC) nude mice model. Materials and Methods: Total of 11 nude mice at 4–6 weeks of age, composed of three groups [tinzaparin treated (n=4), positive control (n=4), negative control (n=3)], were used to create an orthotopic OSCC mice model. Human tongue cancer cell line OSC-19 was utilized to create this in vivo model. After 21 days, the mice were sacrificed and tumors were quantified for BAX and CCNB1 expression utilizing quantitative real-time polimerase chain reaction (qRT-PCR). Livak method normalized for control samples revealed fold-changes utilizing ACTB housekeeping gene. Statistical significance was tested by non-parametric Mann-Whitney U test, where P<0.05 was considered significant. Results: Two mice in positive control and tinzaparin groups did not develop tumors. Available tumors showed decreased BAX and CCNB1 expressions with low statistical significance, when compared to the control group. Conclusion: These preliminary results indicate a deregulated cell cycle control without an effect on apoptosis. More mice should be included to significantly state the effect of tinzaparin treatment in OSCC mice model.

Tinzaparin Tedavisinin Oral Yassı Hücreli Karsinoma Çıplak Fare Modelinde BAX ve CCNB1 Gen Anlatımları Üzerindeki Etkisi

Giriş: Bu çalışma, oral yassı hücreli karsinom (OYHK) çıplak fare modelinde tinzaparin tedavisinin proapoptotik BAX ve hücre döngüsü G2/M faz regülatörü CCNB1 gen anlatım düzeyleri üzerindeki etkisini tespit etmeyi amaçlamıştır. Gereçler ve Yöntemler: Üç gruptan oluşan [tinzaparin tedavisi alan (n=4), pozitif kontrol (n=4), negatif kontrol (n=3)] 4–6 haftalık toplam 11 çıplak farede ortotopik OYHK fare modeli geliştirilmiştir. İnsan dil kanser hücre hattı OSC-19 bu in vivo modelin oluşturulmasında kullanılmıştır. Fareler 21 gün sonra sakrifiye edilerek, tümörler BAX ve CCNB1 gen anlatımları açısından kantitatif eş-zamanlı polimeraz zincir reaksiyonu (kRT-PZR) kullanılarak kantifiye edilmiştir. Gen anlatım düzeyleri Livak metodu ile ACTB housekeeping geni kullanılarak hesaplanmış ve kontrol örneklerine göre normalize edilmiştir. İstatistiksel anlamlılık non-parametrik Mann-Whitney U test ile hesaplanmış ve p değerinin 0.05’den küçük olması anlamlı olarak kabul edilmiştir. Bulgular: Pozitif kontrol ile tinzaparin gruplarından ikişer fare tümör geliştirmemiştir. Geri kalan tümörlerde kontrol grubuna kıyasla azalmış BAX ve CCNB1 gen anlatımı düşük istatistiksel anlamla tespit edilmiştir. Sonuçlar: Bu ön bulgular, apoptozun etkilenmediği, düzeni bozulmuş hücre döngü kontrolünü işaret etmektedir. Daha çok fare ile yapılacak çalışmalar tinzaparin tedavisinin OYHK fare modelindeki anlamlı etkisinin belirlenmesine yardımcı olacaktır.
